摘要 :Molecular interactions between proteins and DNA play crucial roles in many fundamental biological processes, such as DNA modification and gene regulation. DNA-binding proteins encompass various degrees of binding specificity, ranging from highly specific to non-specific. How DNA-binding proteins achieve high binding specificity is still not well understood. In this talk, I will present our recent work on structural features for protein-DNA binding specificity. We investigated both the static and dynamic structural features for achieving high degree of protein-DNA binding specificity. Specifically, we carried out a comparative analysis of protein-DNA complexes with different degrees of binding specificity. We also investigated the role of protein flexibility in protein-DNA binding specificity by comparative molecular dynamics (MD) simulations. Our results show the important roles of protein flexibility and dynamic properties in protein-DNA binding specificity.
个人简介:Dr. Jun-tao Guo is an associate professor in the Department of Bioinformatics and Genomics, College of Computing and Informatics, University of North Carolina at Charlotte. He is also the director of the minor program in bioinformatics. In 2009, he received the prestigious National Science Foundation (NSF) Career Award. His research interest is in the broad area of structural bioinformatics with a special focus on studying molecular interactions. The major research project in his lab is to develop computational methods and resources for modeling protein-DNA interactions and predicting transcription factor binding sites using a structure-based approach on a genomic scale.
